This study evaluated the effects of spirodiclofen, BAJ 2510, and one of the hydroxy BAJ 2510 metabolites, primarily on hormone response elements in human cell lines (MCF 7 breast cancer cell line or prostate PC-3 cell line for estrogen and androgen receptor, respectively). Hormone response was quantified via luciferase reporter genes using a chemiluminescence reader. In some cases, binding studies for alpha and beta estrogen receptors followed, using a fluorescence polarization technique which could be performed in microtiter wells. At pH's of 6 to 6.5, BAJ 2510 displayed estrogenic and anti-estrogenic activities. BAJ 2510 bound to alpha and beta estrogen receptors in this pH range. BAJ 2510 had no effects in the reporter or receptor binding assays at physiological pH (7 or higher). Spirodiclofen and hydroxy BAJ 2510 were uniformly negative in all assays.

Acetogenins, a class of natural compounds produced by some Annonaceae species, are potent inhibitors of mitochondrial electron transport systems. Although the cellular respiration processes are an important biochemical site for the acaricidal action of compounds, few studies have been performed to assess the bioactivity of acetogenin-based biopesticides on spider mites, mainly against species that occur in orchards. Using residual contact bioassays, this study aimed to evaluate the bioactivity of an ethanolic extract from Annona mucosa seeds (ESAM) (Annonaceae) against the citrus red mite Panonychus citri (McGregor) (Acari: Tetranychidae), an important pest of the Brazilian citriculture. The use of synthetic acaricides for management of pest mites may alter the efficacy of the ectoparasitoid Tamarixia radiata (Waterston) in biological control of Diaphorina citri Kuwayama, the vector of the bacteria associated with huanglongbing (HLB) in citrus orchards. We evaluated the toxicity of 16 acaricides that are recommended for the control of citrus-pest mites to T. radiata. Acrinathrin, bifenthrin, carbosulfan, and fenpropathrin caused high acute toxicity and were considered harmful (mortality >77%) to T. radiata. Abamectin, diflubenzuron, etoxazole, fenbutatin oxide, fenpyroximate, flufenoxuron, hexythiazox, propargite, spirodiclofen, and sulfur caused low acute toxicity and affected the parasitism rate and emergence rate of adults (F1 generation), and were considered slightly harmful to T. radiata. Dicofol and pyridaben did not affect the survival and action of the ectoparasitoid, and were considered harmless. In addition to its acute toxicity, carbosulfan caused mortality higher than 25% for >30 d after application, and was considered persistent.

If a grower uses predatory mites, and should use chemical compounds, he needs to be very careful in his choice of products. The selected products have to be efficient against the target pest and at the same time compatible with the present beneficial's. Useful tools for such product selection under greenhouse circumstances are side effects lists. These lists are freely available on the websites of producing companies of biological control agents. But not all products (e.g. newly developed ones) have been tested for side effects. Moreover the information already available in these tables is not based on field tests. For this reason, we have developed a protocol for quick screening of side effects of chemical plant protection products under field conditions. For these experiments we have chosen the predatory mite Amblyseius swirskii as test organism, because this is an often used phytoseiid mite, which is very sensitive to pesticides. Hibiscus rosa sinensis is the standard reference plant in our side effects trials because the chosen predatory mite has shown very good control of pests on this plant species.

A sensitive method for the determination of five new generation pesticides (chlorantraniliprole, spirotetramat, spiromesifen, spirodiclofen and flubendiamide) in soil samples has been developed using liquid chromatography and tandem mass spectrometry (LC-MS(2)) with a triple-quadrupole in the multiple reaction monitoring mode. The target analytes are released from the solid matrix by single-phase extraction in acetonitrile (SLE), this organic phase being used as dispersant solvent in the dispersive liquid-liquid microextraction (DLLME) step. The different parameters affecting the extraction efficiency in DLLME were carefully studied, being 1.5mL of acetonitrile extract (disperser solvent), 125μL carbon tetrachloride (extraction solvent) and 10mL aqueous solution, the selected conditions. The enriched organic phase was evaporated, reconstituted with 50μL acetonitrile and injected into a liquid chromatograph with a mobile phase composed of acetonitrile.
